Pancreatic ductal adenocarcinoma (PDAC) remains one of the most lethal cancers, characterized by a profoundly immunosuppressive tumor microenvironment (TME) and a poor response to current immunotherapies. The TME displays limited T‑cell infiltration and an abundance of regulatory and myeloid cells that dampen anti‑tumor immunity. Our team previously demonstrated that a combined immunotherapy strategy targeting TNFR2 on regulatory T cells, together with CD40 agonist stimulation , can effectively reinvigorate anti‑tumor immune responses in patients. Understanding how treatment shapes the immune landscape and defining predictive biomarkers

will be central to the project through high‑dimensional single‑cell and spatial technologies. This work has strong translational potential for optimizing

immunotherapy strategies for PDAC patients. Where to apply
